2.  Submitting your samples
You can select from three different size standards at this time, depending on the markers you have chosen for your labeled primers.
GS600LIZ – can use markers labeled with 6-FAM (blue), VIC (green), NED (yellow), PET (red).
GS500ROX –can use markers labeled with 6-FAM (blue), HEX (green), NED (yellow).
MM1000ROX—can use markers labeled with 6-FAM (blue), HEX (green), NED (yellow).

Following your PCR, pick up a plate from the Murdock lab, dilute your samples appropriately and aliquot them into the wells.  Each researcher will determine what dilution and aliquot amount to use following optimization.  As a guideline: when using the LIZ600 size, standard, protocol requests .5ul of diluted PCR product per well; and, when using the ROX size standards the request is for 1ul.
Return your plate with diluted product aliquots to the order drop off fridge and notify the Murdock Lab via email that it is in.
Mailing in your plates – we have found the following protocol works well for mailing in plates.

  1. You can order the specific plates (semi-skirted, raised skirt 96 well or 384 well) from any vendor.  Call before purchasing to be sure they will work on the Applied Biosystems 3130xl.  We use the Gold Series Semi-Skirted  96-well pcr plates from ISC-Bioexpress (Bioexpress.com catalog #T-3085-P)—the color allows for better visualization of aliquots.

  2. Aliquot product into wells accordingly.

  3. Spin down the plate briefly to ensure product is seated at bottom of well.

  4. Dry down sample so it does not shift during transit.  One way is to cover the plate lightly with a KimWipe or paper towel to prevent contamination and allow water to evaporate overnight. Please don’t use a Speedvac as this will make it difficult to resuspend.

  5. Seal the plate with a secure plate sealing adhesive cover and ship in a small padded envelope.

  6. Complete the online order form.  Charge cards are preferable, but PO is acceptable.
